چکیده

Single photon counting time distributions obtained from single molecule photon arrival trajectories are employed in the theoretical study of multistate kinetics coupled to a two-state jump bath. The bath time scale may be extracted from statistical analysis of oneand two-point time-domain optical measurements. The amplitude and shape of the distribution of interphoton arrival times reflects bath correlations. A slow bath ~relative to the kinetics! results in broad distributions of arrival times reflecting bath memory. For a fast bath, the arrival time distributions narrow around shorter times, similar to motional narrowing in frequency domain spectroscopy. The variance of either kinetic rates or equilibrium population of bath states results in asymmetry of the distribution of two photon arrival times. © 2002 American Institute of Physics. @DOI: 10.1063/1.1515321#

Fluorescence correlation spectroscopy as a tool to investigate single molecule probe dynamics in thin polymer films.

Fluorescence correlation experiments were performed on rhodamine 6G in PDMS spin-coated films on glass surfaces. With polarised excitation, ensemble bleaching of the dye and single molecule intensity fluctuations were observed. From the statistics of single molecule intensity data taken at different positions in the film, correlation functions were calculated. Two modes of motion with exponenti...

Probing Protein Multidimensional Conformational Fluctuations by Single-Molecule Multiparameter Photon Stamping Spectroscopy

Conformational motions of proteins are highly dynamic and intrinsically complex. To capture the temporal and spatial complexity of conformational motions and further to understand their roles in protein functions, an attempt is made to probe multidimensional conformational dynamics of proteins besides the typical one-dimensional FRET coordinate or the projected conformational motions on the one...

Theory of single-molecule spectroscopy: beyond the ensemble average.

Single-molecule spectroscopy (SMS) is a powerful experimental technique used to investigate a wide range of physical, chemical, and biophysical phenomena. The merit of SMS is that it does not require ensemble averaging, which is found in standard spectroscopic techniques. Thus SMS yields insight into complex fluctuation phenomena that cannot be observed using standard ensemble techniques. We in...

Structural Analysis of Hirudin Using FT-IR and FT-Raman Spectroscopic Techniques

FT-IR and FT-Raman spectra of hirudin have been recorded from the native solid hirudin. The conformation of the molecule has been discussed on the basis of IR and Raman data. It has been concluded that hirudin molecule has a mixed a-helix and random coil conformation.

Internally labeled Cy3/Cy5 DNA constructs show greatly enhanced photo-stability in single-molecule FRET experiments

DNA constructs labeled with cyanine fluorescent dyes are important substrates for single-molecule (sm) studies of the functional activity of protein-DNA complexes. We previously studied the local DNA backbone fluctuations of replication fork and primer-template DNA constructs labeled with Cy3/Cy5 donor-acceptor Förster resonance energy transfer (FRET) chromophore pairs and showed that, contrary...
